Abstract
Eucalyptus L'Herit. is a large economically and ecologically important genus of over 700 species, endemic to Australia and several tropical islands to the north of Australia. Taxonomic subdivision of the genus is difficult because of high levels of quantitative variation and homoplasy among morphological characters, low levels of phylogenetically useful variation in ribosomal and chloroplast DNA, and incomplete reproductive isolation of species that results in interspecific hybridisation and introgression. A newly developed set of DArT (Diversity Arrays Technology) markers are heralding a new era for Eucalyptus phylogenetics based on high-resolution, genome-wide comparisons. DArT, originally developed for use in population genetics, association genetics and genome mapping, is proving to be highly informative about relationships at all taxonomic levels within Eucalyptus. We have developed a large set of DArT markers sourced from eight eucalypt species and these have proven to be highly transferable between species within the genus. However, DArT markers developed from Corymbia, a genus that was until recently included within Eucalyptus, were not transferable to Eucalyptus, and vice versa. Phylogenetic analysis of data from over 8000 DArT markers resolves relationships between subgenera, sections and species of Eucalyptus. The congruence of the results with those from ITS sequence data, AFLP data and traditional morphology-based classifications gives us confidence that genome-wide genotyping with DArT will allow rapid and relatively inexpensive resolution of previously intractable phylogenetic issues within this globally important genus.
