- Parchment is a collagen-based, historically important biomaterials that contains many layers of information, from text written on the surface, to the structure of the material itself. The degradation of historical parchments is often attributed to inappropriate storage conditions, although other factors may also accelerate the decay of collagen within the parchment, such as harsh cleaning or manufacturing techniques that involved extreme variations in pH or mechanical treatment. X-ray diffraction at small and wide angles is an ideal tool for analysing the structure of the collagen, which gives the parchment its strength and durability over time. This chapter describes how small angle X-ray scattering (SAXS), in conjunction with sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and thermal techniques, and wide angle X-ray diffraction (WAXD) can give a detailed account of the collagen structure within the parchment and indicate any deterioration brought about by laser-cleaning. © 2006 Elsevier B.V.
